Humatrope® is a polypeptide hormone derived from recombinant DNA. It has a molecular weight of approximately 22,125 daltons and consists of 191 amino acid residues. The amino acid sequence of Humatrope is identical to that of human growth hormone produced by the pituitary gland. The hormone is synthesized in Escherichia coli bacteria modified with the human growth hormone gene.
Humatrope is a sterile, lyophilized powder available for subcutaneous or intramuscular administration after reconstitution. It is a highly purified preparation, and the pH of the reconstituted solution is approximately 7.5. It is important to note that Humatrope is sensitive to oxygen.
The vials of Humatrope contain 5 mg of somatropin (equivalent to 15 IU or 225 nanomoles), along with mannitol (25 mg), glycine (5 mg), and dibasic sodium phosphate (1.13 mg). Each vial is accompanied by a 5-mL vial of diluting solution containing water for injection, 0.3% Metacresol as a preservative, and 1.7% glycerin.
For cartridge presentations, the somatropin content varies. The cartridges contain either 6 mg (18 IU), 12 mg (36 IU), or 24 mg (72 IU) of somatropin. Along with somatropin, the cartridges also contain mannitol, glycine, and dibasic sodium phosphate. The diluting solution in the cartridge presentation includes water for injection, 0.3% Metacresol as a preservative, and varying amounts of glycerin.
It stimulates linear growth in pediatric patients by targeting the growth plates of long bones and increasing concentrations of insulin-like growth factor-I (IGF-I). Humatrope also promotes skeletal muscle growth by increasing the number and size of muscle cells. It facilitates protein synthesis and nitrogen retention, which contribute to the overall growth process.
